Relapsedandrefractorydiseaseshortensthesurvivalofacutemyeloidleukemia(AML)patients.Therefore,additional therapyapartfromconventionalchemotherapyandstemcelltransplantationisurgentlyneeded,especiallyineradicatingresidual leukemic stem cells. The identiï¬cation of leukemia associated antigens, and the observation that administration of allogeneic T cellsmaymediategraftversusleukemiaeï¬ectpavedthewaytodevelopvariousimmunotherapystrategies.Eï¬ectiveprovisionof bothtumorantigenandco-stimulationareessentialforstrategiesaimedatenrollingcytotoxicT-lymphocytestoeradicateleukemic cells.Inthisstudy,weattemptedtoup-regulateco-stimulatorsonleukemiccells(termed“dendritisation”tomimicdendriticcells inprovidingstrongco-stimulation)andcouplewithinherentleukemicantigenstogeneratespeciï¬cantileukemicT-lymphocytes againstautologousblastcells.OurstudyshowsevidenceofdistinctcytotoxicityofCD4positiveTlymphocytesagainstautologous blastcells.DendritisedAMLblastswereabletofunctionwithantigenpresentingcapability,whilestillphenotypicallyexistingas blastcells.ThiswasalsoachievableforawiderangeofsubtypesofAML,makingitapotentialimmunotherapyagainstAML
